Itraconazole is an antifungal agent that belongs to the class of imidazoles. It works by inhibiting the cell wall synthesis in fungi. Itraconazole inhibits the activity of the enzyme, cytochrome P450 14 a-demethylase, which is involved in the conversion of lanosterol to ergosterol. When the synthesis of ergosterol is inhibited, it leads to increased cellular permeability, leakage of the cellular components and eventually leads to cell death. Other mechanisms by which itraconazole inhibits the fungal growth include inhibition of endogenous respiration, purine uptake inhibition and impaired phospholipid biosynthesis.

This medication is contraindicated in patients with congestive heart failure, onychomycosis and those with liver and kidney impairment.
Adverse effects of this medication include hepatotoxicity, headache, constipation, nausea, fever, vomiting dizziness and upset stomach.
There are no sufficient and well-controlled studies when it comes to itraconazole administration in pregnant women and nursing mothers. Therefore, if you are pregnant or nursing, discuss with your doctor about your condition before taking this medicine.
