Each 5 ml contains

  • Ambroxol HCl 15 mg
  • Guaiphenisin 50 mg
  • Phenylephrine HCl 5 mg
  • Chlorpheniramine maleate 2 mg
  • Menthol 1 mg


Ambroxol is a clinically proven systemically active mucolytic agent. When administered orally onset of action occurs after about 30 minutes. The breakdown of acid mucopolysaccharide fibers makes the sputum thinner and less viscous and therefore more easily removed by coughing. Although sputum volume eventually decreases, its viscosity remains low for as long as treatment is maintained. Ambroxol increases content of surfactant in lungs, which is dealt with strengthening of synthesis of the last and secretion in alveolar pneumocytes, and also with breach of its disintegration. The medicinal product increases mucociliar transport of phlegm. It suppresses coughing insignificantly.

Guaiphenesin has an expectorant action. It reduces sputum viscosity by increasing the volume and water content of the bronchial secretion thereby facilitating the expectoration of sputum.

Chlorpheniramine maleate is a first generation antihistamine which provides prompt relief of itchy-watery eyes, runny nose, sneezing, itching of the nose or throat due to respiratory allergies. The effectiveness of first-generation antihistamines in blocking sneezing in colds may be due primarily to neuropharmacological manipulation of histaminic and muscarinic receptors in the medulla.

Phenylephrine is a sympathomimetic vasoconstrictor that has been used as a nasal decongestant for many years. Phenylephrine constricts the blood vessels in the nasal mucous membranes and allows the air passages to open up. It is a relatively selective alpha-adrenoceptor agonist. The majority of the sympathomimetic action is due to direct stimulation of the adrenoceptors. At therapeutic doses, it does not cause significant stimulation of the central nervous system.

Menthol has a cooling effect on the throat. It has been suggested that benefits of menthol may be due to an effect on calcium channels of sensory nerves.


PENTAKOF-PC Syrup is indicated for the treatment of upper respiratory tract infections associated with cough ,cold ,fever and body ache.

The use of PENTAKOF-PC Syrup Tablet is contraindicated in patients with:

  • Hypersensitivity to any of the ingredients of the formulation.
  • Severe hypertension.


In case a hypersensitivity reaction occurs which is rare, PENTAKOF-PC Syrup should be discontinued.

PENTAKOF-PC Syrup should be used with caution in patients with renal or hepatic dysfunction, diabetes mellitus, hyperthyroidism, cardiovascular problems, epilepsy and closed angle glaucoma. It is advisable not to drive or operate machinery when on treatment with PENTAKOF-PC Syrup.

Concurrent administration of CNS depressants may have additive effect leading to sedation /somnolence. .


PENTAKOF-PC is generally well tolerated and adverse events are rare. Hypersensitive individuals may display ephedrine-like reactions such as tachycardia, palpitations, headache, dizziness and nausea. Use of sympathomimetics has been associated with fear, anxiety, restlessness, tremor, weakness, dysuria, insomnia, hallucinations and convulsions. Chlorpheniramine may cause sedation. Ambroxol may cause abdominal discomfort and hypersensivity reactions. Guaiphenesin may cause gastrointestinal discomfort, nausea and vomiting. is generally well tolerated and adverse events are rare.


For the safety in pregnancy and lactation,probable benefits should be weighed against possible risk ,before prescribing.
